Composition
Methotrexate 10 mg
Description
The active substance of Prestrex 10 tablets is methotrexate, an ant metabolite and immunosuppressant (medicine which affects the reproduction of the body’s cells and reduces the activity of the immune system) Methotrexate 10 MG Tablet is used in the treatment of life-threatening conditions like rheumatoid arthritis, psoriasis.
Indications
- Severe resistant disabling psoriasis
- Severe psoriatic arthritis in adult patients
- Active rheumatoid arthritis in adult patients
Dosage
Dosage for rheumatoid arthritis, psoriasis and severe psoriatic arthritis, The recommended dose is 7.5 – 15 mg orally, once weekly.
This should be adjusted according to your response to treatment and side effects. Your doctor may also instruct you to take the weekly dose in three divided doses over 24 or 36 hours.
Mechanism of Action
For rheumatoid arthritis, it works by reducing inflammation and by suppressing the immune system. For psoriasis, methotrexate works by attacking the rapidly dividing cells that are involved in causing psoriasis. Under these circumstances, methotrexate is known as a disease-modifying antirheumatic drug (DMARD)

Absorption
oral (low doses only) readily absorbed from GI tract, high bioavailabilitypeak plasma concentrations may occur 1 to 5 hours after a dose of 15 to 30 mg/m2. it is suggested that the drug be taken on an empty stomach with clear liquids.
Distribution
After intravenous (IV) administration, MTX distributes within an initial volume of approximately 18% (0.18 L/kg of body weight). The steady-state volume is 40% to 80% of body weight. The initial distribution phase has a t1/2 of 30 to 45 min
Metabolism
The major metabolite of MTX, produced by the action of hepatic aldehyde oxidase, is 7-hydroxy MTX (7-OH-MTX). 7-OH-MTX is only 1% as potent an inhibitor of DHFR as is MTX.It is also less water soluble than is MTX and may contribute to the renal toxicity frequently seen after high doses of the antifolate.

- Diabetes mellitus treated with insulin.
